Do you still need to disconnect the negative cable form the Aux battery and pull fuse 42 if you install the Auto Stop/Start Eliminator?

Think of disabling ESS as a two-step procedure:

1. Isolating the ESS battery, via disconnecting the related Negative cable and removing Fuse F42. Doing so will prevent that battery from eventually cannibalizing the vehicle's main 12V battery. However, this won't prevent ESS from functioning; so long as it isn't turned off, restarts will pivot to the main battery for power.

2. Using a defeat device, like the Auto Start/Stop Eliminator, creates a memory function. If the driver turns ESS off, the harness will remember that setting after the vehicle is shut down and restarted.

If you have decided to turn off ESS events from happening there is in my opinion nothing but advantage to disconnecting the ESS/Aux battery and pulling the aforementioned Fuse to prevent it from being isolated.

The ever so slight advantage of keeping it connected, in additional battery power, is enormously outweighed, I think, by the potential for this battery to not only go bad, but in so doing risk taking the main battery out with if in remains connected.

It serves little purpose but to keep appliances running during the very ESS events you've chosen to bypass.

LOL you’re like the girl at a party that thinks all the other girls are talking about her when they’re not. If it makes you feel better, I meant it as a compliment to @Heimkehr who is clear and concise and easy for me to follow. If my teasing from time to time hurts your feelings I apologize.
